In reaching its decision on MRLs for danofloxacin, the Committeetook into account the following: - An ADI of 0-20 µg per kg of body weight was established, based ona toxicological end-point. This would result in a maximum ADI of1200 µg for a 60-kg person. - The parent drug is the marker residue. - The metabolite desmethyldanofloxacin is considered to be tentimes as toxic as the parent drug. - Only the toxicological activity of the parent drug and desmethyl-danofloxacin are known. Therefore all the residues, including thebound residues, are assumed to be of toxicological concern. The un-identified residues are assumed to have the same toxicity as theparent drug. - Liver and muscle were considered to be the appropriate targettissues. - Desmethyldanofloxacin occurs only in the liver. - The proportion of the total residues accounted for by danofloxacin and desmethyldanofloxacin varies between different species. The sum of all the residues covered by the AD1 expressed asdanofloxacin equivalents is given by the formula: Total residues = residues of danofloxacin + unidentified residuesexpressed as danofloxacin equivalents + residuesof desmethyldanofloxacin expressed as danofloxacinequivalents.
